Profiling Endogenous Opioid Peptide Release from Adrenal Chromaffin Cells
Adrenal chromaffin cells regulate the response to acute stressors by releasing catecholamines and a range of other molecules, including opioid peptides derived from pro-enkephalin. Fast-scan cyclic voltammetry can be used to codetect the exocytotic release of catecholamine and enkephalin peptides from individual bovine adrenal chromaffin cells, revealing differential release kinetics for these classes of molecules. A deep investigation into the identity of the released neuropeptides via liquid chromatography-tandem mass spectrometry profiles multiple variants of the enkephalin peptides released, emphasizing the biological complexity of opioid peptide signaling.
